Thymic stromal lymphopoietin (TSLP) is an epithelial-derived "alarmin" cytokine positioned at the very top of the allergic cascade. Triggered by barrier damage, allergens, viruses and pollutants, TSLP conditions dendritic cells toward a Th2-priming phenotype and acts directly on innate lymphoid cells and basophils, initiating the type 2 inflammation that underlies atopic dermatitis, allergic asthma and chronic rhinosinusitis with nasal polyps. Blocking TSLP with tezepelumab — the first upstream biologic for severe asthma — validated the target clinically and ignited a pipeline of TSLP-directed antibodies and small molecules. Recombinant human TSLP supports receptor-binding assays, anti-TSLP drug screening and epithelial immunology research.
