Bioactivity
Measured by functional ELISA. Immobilized recombinant human IL-17A binds a reference anti-IL-17A antibody (secukinumab/ixekizumab class) with an EC50 in the range of 20–80 ng/mL.
Background
Interleukin-17A is the signature effector cytokine of Th17 cells and the founding member of the IL-17 family. Signaling as a disulfide-linked homodimer through the heteromeric IL-17RA/IL-17RC receptor complex, IL-17A drives epithelial and stromal production of neutrophil-attracting chemokines, antimicrobial peptides and inflammatory mediators — defending mucosal barriers against extracellular fungi and bacteria, but also igniting the pathology of psoriasis, psoriatic arthritis and axial spondyloarthritis. Blocking IL-17A or its receptor with secukinumab, ixekizumab or brodalumab delivers dramatic clinical responses in these diseases. Recombinant human IL-17A supports anti-cytokine antibody screening, receptor-binding and neutralization assays, and Th17-axis research.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
