Bioactivity
Measured by functional ELISA. Immobilized recombinant human IL-23 binds reference anti-p19 antibodies (guselkumab/risankizumab class) with an EC50 in the range of 20–80 ng/mL.
Background
Interleukin-23 is a disulfide-linked heterodimeric cytokine composed of the IL-23-specific p19 subunit (IL23A) and the p40 subunit (IL12B) shared with IL-12. Acting through the IL-23R/IL-12Rβ1 receptor complex on Th17 cells, innate lymphoid cells and γδ T cells, IL-23 maintains and expands the IL-17-producing effector population that drives chronic inflammation in psoriasis, IBD and other autoimmune settings — the upstream "IL-23/IL-17 axis". Therapeutically, p19-selective antibodies such as guselkumab and risankizumab, and the dual p40 blocker ustekinumab, have become mainstays of dermatology and gastroenterology. Bioactive recombinant IL-23 heterodimer is essential for pathway screening, neutralization assays and Th17 biology.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
