Integrin alpha-4 beta-7 is the gut-homing integrin: on memory lymphocytes it recognizes MAdCAM-1 on intestinal endothelium (and fibronectin), directing cells to mucosal immune sites. Its clinical significance was established by vedolizumab (Entyvio), an anti-β7 antibody that selectively blocks gut recruitment while leaving systemic immunity intact — the prototype of site-targeted immunotherapy in ulcerative colitis and Crohn's disease. New α4β7-directed molecules and gut-selective programs continue to build on this axis. The mammalian-expressed α4β7 ectodomain heterodimer preserves the conformational ligand-binding site required for authentic MAdCAM-1 binding and antibody-blocking studies.
