Bioactivity
Measured by functional ELISA. Immobilized recombinant human PCSK9 binds recombinant human LDLR and reference anti-PCSK9 antibody (evolocumab/alirocumab class) with EC50 values in the ng/mL range.
Background
PCSK9 (proprotein convertase subtilisin/kexin type 9) is a secreted serine protease that has become the second most important lever on LDL cholesterol after statins. Secreted by the liver, PCSK9 binds the LDL receptor on hepatocytes and escorts it to lysosomal degradation; blocking this interaction with the antibodies evolocumab and alirocumab, or silencing PCSK9 with inclisiran, dramatically lowers LDL-C and cardiovascular risk. The secreted protein is processed from a zymogen into a prodomain plus mature catalytic form that remains non-covalently associated — both parts participating in receptor binding. Recombinant human PCSK9 is the standard reagent for PCSK9–LDLR competition assays, antibody screening and lipid-metabolism research.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
