RANKL (receptor activator of NF-κB ligand, TNFSF11) is the TNF-family cytokine that commands osteoclast differentiation and activation, coupling bone resorption to immune and endocrine signals through its receptors RANK and the decoy OPG. Excessive RANKL drives osteoporosis, bone metastases and myeloma bone disease — pathology systemically silenced by the anti-RANKL antibody denosumab. Like other TNF-family ligands, RANKL is biologically active only as a homotrimer, which presents the receptor-binding interfaces in native geometry. Our MALS-verified trimeric RANKL supports denosumab-class biosimilar QC, RANK blocking assays and osteoclastogenesis research.
