Bioactivity
Measured by functional ELISA. Immobilized recombinant human CD79B binds polatuzumab vedotin-derived reference antibody with an EC50 in the range of 10–50 ng/mL.
Background
CD79B (Igβ, B29) is the signaling subunit of the B-cell receptor: paired with CD79A, it associates with surface immunoglobulin and delivers activation signals through its cytoplasmic ITAM motifs. Because CD79B is expressed across the B-cell lineage — including malignant B cells that may lose surface immunoglobulin expression — and is efficiently internalized upon antibody binding, it has become a premier target for antibody–drug conjugates in lymphoma. Polatuzumab vedotin established the target clinically in diffuse large B-cell lymphoma, and next-generation CD79B ADCs and bispecifics are advancing rapidly. The extracellular Ig-like domain presented here is the epitope-bearing region accessible to therapeutic antibodies, supporting ADC screening, affinity maturation and potency assay development for the growing CD79B pipeline.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
