Bioactivity
Measured by functional ELISA. Immobilized recombinant human CD38 binds reference anti-CD38 antibodies daratumumab and isatuximab with EC50 values in the range of 10–50 ng/mL.
Background
CD38 is a type II transmembrane ectoenzyme whose extracellular domain catalyzes the synthesis and hydrolysis of cyclic ADP-ribose, linking surface receptor signaling to intracellular calcium mobilization and NAD+ metabolism. As a marker, CD38 is expressed at high density on plasma cells, activated T and NK cells and myeloid cells, and its uniform, intense expression on multiple myeloma cells made it an outstanding therapeutic target: the CD38 antibodies daratumumab and isatuximab achieve deep responses in myeloma through Fc-mediated effector mechanisms and immune modulation. CD38 is also a rising target in autoimmune disease and sickle-cell research. The soluble extracellular domain presented here supports anti-CD38 antibody screening, biosimilar potency testing, enzyme-activity studies and diagnostic assay development.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
