Bioactivity
Measured by functional ELISA. Immobilized recombinant human CD30 binds brentuximab vedotin-derived reference antibody and soluble CD30L (TNFSF8) with EC50 values in the ng/mL range.
Background
CD30 (TNFRSF8) is a member of the tumor necrosis factor receptor superfamily and a classic marker of lymphoid activation. Although low in most resting lymphocytes, CD30 is abundantly expressed on Hodgkin/Reed–Sternberg cells and anaplastic large-cell lymphoma, and its restricted expression profile made it the model antigen for antibody–drug conjugates: brentuximab vedotin pairs an anti-CD30 antibody with a microtubule-disrupting payload and has become standard therapy in Hodgkin lymphoma and ALCL. Soluble CD30 released by shedding serves as a disease-monitoring biomarker. Beyond ADCs, CD30-directed CAR-T constructs and bispecifics are under active development. Recombinant human CD30 extracellular domain provides the full ligand- and antibody-binding ectodomain needed for ADC screening, biosimilar QC and ligand–receptor studies.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
