Bioactivity
Measured by functional ELISA. Immobilized recombinant human IL-6R binds IL-6 and reference anti-IL-6R antibody (tocilizumab) with EC50 values in the ng/mL range.
Background
The interleukin-6 receptor alpha chain (IL-6R, CD126) initiates both classic membrane-bound signaling and — after proteolytic shedding or alternative splicing to soluble IL-6R — the trans-signaling program that renders virtually every cell IL-6 responsive. IL-6 drives the acute-phase response, B-cell maturation and Th17 differentiation, and its chronic elevation underlies rheumatoid arthritis, giant cell arteritis and the cytokine storm of severe inflammation. Tocilizumab and sarilumab block the receptor and are mainstay biologics in rheumatology; soluble IL-6R levels stratify patient response. Recombinant IL-6R extracellular domain supports ligand-capture assays, anti-IL-6R biosimilar testing and trans-signaling research.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
