Bioactivity
Measured by functional ELISA. Immobilized recombinant human GLP-1R ECD binds GLP-1(7-36) amide and reference GLP-1R-targeting peptides with EC50 values in the ng/mL range.
Background
The glucagon-like peptide-1 receptor (GLP-1R) is a class B GPCR that translates incretin signaling into glucose-dependent insulin secretion, appetite suppression and cardiovascular benefit — the pharmacology behind the semaglutide, liraglutide and tirzepatide therapeutic revolution in type 2 diabetes and obesity. Its extracellular ligand-binding domain is the primary determinant of peptide-agonist recognition, capturing the C-terminal helix of GLP-1 and engineered analogs before membrane-domain activation. The dimeric hFc fusion presented here stabilizes the ECD for plate-based and label-free binding assays, supporting analog screening, affinity ranking and pharmacodynamic comparator testing across the incretin development landscape.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
