Bioactivity
Measured by functional ELISA. Immobilized recombinant human PD-L2 binds recombinant human PD-1 with an EC50 in the range of 10–50 ng/mL; the interaction is blocked by reference anti-PD-L2 antibodies.
Background
PD-L2 (programmed death-ligand 2), also known as B7-DC and PDCD1LG2, is the second ligand of the PD-1 receptor and a member of the B7 immunoglobulin superfamily. In contrast to the broadly expressed PD-L1, PD-L2 expression is largely restricted to antigen-presenting cells — macrophages and dendritic cells — and is strongly induced during Th2-type immune responses. PD-L2 binds PD-1 with two- to three-fold higher affinity than PD-L1 and contributes to immune regulation in the lung, gut and other barrier tissues, with documented roles in asthma, infection and tumor immune evasion that are increasingly dissected separately from the PD-L1 axis. Because therapeutic antibodies and bispecifics can differ sharply in their PD-L1 versus PD-L2 cross-reactivity, recombinant human PD-L2 is a key reagent for selectivity profiling, ligand–receptor blocking assays and checkpoint pipeline development.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
