Bioactivity
Measured by functional ELISA. Immobilized recombinant human DLL3 binds reference anti-DLL3 antibody (tarlatamab class) with an EC50 in the range of 10–50 ng/mL.
Background
DLL3 (Delta-like canonical Notch ligand 3) is an atypical Notch-family ligand that, uniquely among its relatives, inhibits canonical Notch signaling and is sequestered intracellularly in most tissues. In neuroendocrine tumors — above all small-cell lung cancer — DLL3 re-emerges on the cell surface at high density with minimal normal-tissue expression, making it an almost ideal tumor-restricted target. That biology underwrote tarlatamab, the first DLL3×CD3 bispecific T-cell engager approved for SCLC, alongside earlier rovalpituzumab tesirine ADC learnings. The full extracellular domain spanning the DSL and EGF-like repeats presented here supports bispecific screening, potency assay development and DLL3 diagnostic reagent work.
Formulation & Handling
- Formulation
- Supplied in either a lyophilized powder or a ready-to-use liquid format. Buffer: PBS, pH 7.4, with 5% trehalose as cryoprotectant.
- Reconstitution
- Lyophilized: briefly centrifuge the vial before opening, reconstitute in sterile distilled water to 0.1–1.0 mg/mL, and let stand at room temperature for 10 minutes before aliquoting. Liquid: ready to use — thaw on ice and equilibrate.
- Storage
- Both lyophilized and liquid proteins are stable for 12 months at -40 °C to -80 °C. After reconstitution, aliquot and store at -40 °C to -80 °C. Avoid repeated freeze-thaw cycles.
- Shipping
- Shipped on dry ice or cold packs.
